目的探讨卵巢癌基因HE4对卵巢癌细胞增殖及侵袭能力的影响。方法构建针对HE4基因的PcDNA3.1/HE4载体,脂质体法介导PcDNA3.1/HE4载体转染人浆液性卵巢癌细胞系HO8910,WESTERN-BLOT检测转染前后细胞内HE4蛋白的表达效果。用四甲基偶氮唑蓝(MTT)比色法、平板克隆形成实验观察HE4基因对细胞增殖能力的影响,经细胞粘附实验、体外侵袭力实验比较转染前后细胞侵袭能力的变化。结果Western-blot结果显示PcDNA3.1/HE4载体成功转染HO8910细胞并显著提高该细胞HE4基因的表达,细胞增殖能力较前无明显变化,侵袭能力明显下降。结论通过增强HE4基因表达,可降低其侵袭能力,有望成为卵巢癌基因治疗的新的候选基因。
Objective To investigate the effect of ovarian cancer gene HE4 on the proliferation and invasion of ovarian cancer cells. Methods The pcDNA3.1 / HE4 vector targeting HE4 gene was constructed and transfected into human ovarian carcinoma cell line HO8910 by pcDNA3.1 / HE4 vector by liposome method. The expression of HE4 protein was detected by WESTERN-BLOT . The effect of HE4 gene on cell proliferation was observed by MTT colorimetric assay and plate clone formation assay. The cell invasion ability was compared by cell adhesion assay and in vitro invasiveness assay. Results Western-blot results showed that pcDNA3.1 / HE4 vector was successfully transfected into HO8910 cells and significantly increased the expression of HE4 gene in this cell line. The cell proliferation ability was not significantly changed compared with the former, and the invasion ability was significantly decreased. Conclusion HE4 gene expression can reduce the invasiveness of HE4 gene and may be a new candidate gene for ovarian cancer gene therapy.
