目的以果蝇为对象研究山楂提取物(hawthorn fruit extract,HFE)延长寿命和抗氧化的作用。方法收集2d龄的雄性和雌性果蝇,平均分为4组,饲喂添加不同浓度(0、0.5、1.5、4.5mg/ml)山楂提取物的培养基。研究HFE对果蝇生存寿命,超氧化物歧化酶(SOD)、过氧化氢酶(CAT)等抗氧化酶活力及抗氧化相关基因表达的影响;用急性实验研究HFE对果蝇生存时间的影响。结果与对照组相比,1.5、4.5mg/ml HFE能够明显改善果蝇的抗氧化能力,果蝇体内SOD、CAT活力较空白对照组都有显著增加,丙二醛(MDA)含量显著下降;4.5mg/ml HFE组果蝇体内SOD、CAT m RNA表达水平显著上调,雄蝇methuselah(MTH)呈下调趋势;急性实验中HFE组果蝇的寿命明显延长。结论 HFE在体内具有较强的抗氧化活性,并可能通过上调抗氧化基因表达实现延长果蝇寿命的作用。
Objective To study the effect of hawthorn fruit extract (HFE) on prolonging life and antioxidation of fruit flies. Methods Male and female Drosophila melanogaster of 2 days old were collected and equally divided into 4 groups and fed with different concentrations of hawthorn extract (0, 0.5, 1.5, 4.5mg / ml). To study the effect of HFE on the life span, the activities of antioxidant enzymes such as superoxide dismutase (SOD) and catalase (CAT) and the expression of antioxidant genes in Drosophila melanogaster. Acute experiments were conducted to study the effect of HFE on the survival time of Drosophila . Results Compared with the control group, HFE at 1.5 and 4.5 mg / ml significantly improved the antioxidant capacity of Drosophila melanogaster. Compared with the control group, the activities of SOD and CAT in Drosophila melanogaster increased significantly and the content of malondialdehyde (MDA) decreased significantly. The levels of SOD and CAT m RNA in Drosophila melanogaster at 4.5 mg / ml group were significantly up-regulated, while the methuselah (MTH) at male flies showed a downward trend. In the acute experiment, the life span of Drosophila melanogaster was prolonged significantly. Conclusion HFE has strong antioxidant activity in vivo and may extend the lifespan of Drosophila by up-regulating the expression of antioxidant genes.
