目的通过正交设计优化CTL细胞最佳杀瘤条件。方法补体裂解法分离CTL细胞,MTT法测定CTL细胞杀瘤活性,正交设计选择CTL细胞最佳杀瘤条件。结果在SEB浓度为10-6g/ml,SEB作用CTL细胞时间为24h,CTL细胞与肿瘤细胞之比为20∶1情况下CTL细胞杀瘤活性最强。结论CTL细胞最佳杀瘤条件组合为SEB10-6g/ml,作用时间24h,效靶比20∶1。
Objective To optimize the optimal killing conditions of CTL cells by orthogonal design. Methods CTL cells were isolated by complement lysis assay. The cytotoxic activity of CTL cells was measured by MTT assay. The optimal killing conditions of CTL cells were selected by orthogonal design. Results The anti-tumor activity of CTL cells was the strongest when the ratio of CTL cells to tumor cells was 20:1 when the concentration of SEB was 10-6 g/ml and the time of CTL cells treated with SEB was 24 h. Conclusion The optimal killing conditions of CTL cells were SEB10-6g/ml, the action time was 24h, and the effective target ratio was 20∶1.
