目的: 探讨草棉花提取物聚Ⅰ和聚Ⅱ对ADP和胶原诱导的大鼠血小板聚集功能的影响。方法: 以二磷酸腺苷(Adenosine 5′Diphosphate,ADP,终浓度为3.33μmol/L)和胶原(Collagen,终浓度为212μg/L)诱导血小板聚集,聚Ⅰ和聚Ⅱ分别分为25.0、50.0、100.0 mg/kg 3个剂量组,阿司匹林(17.1 mg/kg, ig)和丹参滴丸(72.9mg/kg,ig)为阳性对照,记录5 min内聚集曲线,以最大聚集率和抑制率为观察指标。结果: 聚Ⅰ(50.0、100.0 mg/kg, ig)、聚Ⅱ(50.0、100.0 mg/kg, ig)对ADP诱导的大鼠血小板聚集有明显抑制作用(P<0.05或0.01),抑制率分别为16.79%、21.98%和11.71%、17.67%,呈剂量依赖性。聚Ⅰ(100.0 mg/kg, ig)对Col诱导的血小板聚集有明显抑制作用(P<0.05),抑制率为13.35%。结论: 聚Ⅰ对血小板聚集抑制作用较聚Ⅱ强,可在确定聚Ⅰ提取工艺的基础上进一步优化。
Objective: To investigate the effects of poly-I and poly-II on the platelet aggregation of rats induced by ADP and collagen. Methods: Platelet aggregation was induced by adenosine diphosphate (Adenosine 5′Diphosphate, ADP, final concentration of 3.33 μmol/L) and collagen (Collagen, final concentration of 212 μg/L). Poly I and poly II were divided into 25.0 and 50.0, respectively. In the 100.0 mg/kg 3 dose group, aspirin (17.1 mg/kg, ig) and Danshen Dripping Pills (72.9 mg/kg, ig) were positive controls. The accumulation curve was recorded within 5 min, with the maximum aggregation rate and inhibition rate. Observe the indicators. Results: Poly(I, 50.0, 100.0 mg/kg, ig), Poly II (50.0, 100.0 mg/kg, ig) significantly inhibited ADP-induced platelet aggregation in rats (P<0.05 or 0.01), and the inhibition rates were 16.79%, 21.98% and 11.71%, 17.67%, in a dose-dependent manner. Poly I (100.0 mg/kg, ig) significantly inhibited Col-induced platelet aggregation (P<0.05), and the inhibition rate was 13.35%. Conclusion: Poly I has stronger inhibition on platelet aggregation than Poly II. It can be further optimized based on the determination of poly I extraction process.
