应用噬菌体表面递呈表达系统构建了人抗体组合文库,并筛选出了结合乙肝表面抗原(HBsAg)的人噬菌体抗体(Fab片段)。以免疫球蛋白信号肽序列为引物进行半套式PCR所得到的产物在质和量上都优越于以可变区5′末端保守序列为引物进行PCR所得到的产物。经过3次亲和选择后,抗体阳性率为69%。抑制实验表明,所筛选出来的噬菌体抗体具有抗HBsAg的特异性。序列分析表明VH分别属于VHI亚群和Ⅲ亚群,其轻链VL分别属于VλⅡ亚群和VλⅠ亚群。
The phage display system was used to construct the human antibody combinatorial library, and the human phage antibody (Fab fragment) bound to hepatitis B surface antigen (HBsAg) was screened out. The product obtained by semi-nested PCR using the immunoglobulin signal peptide sequence as a primer is superior in quality and quantity to the product obtained by PCR using the 5 ’end conservative sequence as a primer. After three affinity selection, the antibody positive rate was 69%. Inhibition experiments showed that the selected phage antibodies have anti-HBsAg specificity. Sequence analysis showed that VH belonged to VHI subgroup and subgroup III respectively, and light chain VL belonged to VλⅡ subgroup and VλⅠ subgroup respectively.
