本工作与福建省热作所生理组协作,在室内零上低温条件下,对不同抗寒性品系的胶苗细胞结构形态进行显微观察,着重探讨细胞核和叶绿体结构形态稳定性与抗寒性的关系.实验采用三种类型(根据本省寒害评级资料),即抗寒性强的天任31—45、东试93—114、五凤1、闽林71—22,抗寒性中等的海垦2、五星Ⅰ_3、GT_1以及抗寒性弱的PP86、PR107、RRlM600共十个品系,取顶芽稳定实生苗离体15cm左右枝条及实验系比区无性系顶蓬稳定叶片为材料,在冰箱中4—5℃进行低温处理,分别作成活体制片,配合中性红、地衣红染色,显微观察其苗茎和叶片表皮细胞核以及皮层细胞叶绿体,并进行显微摄影.初步结果如下:
