基于血脂代谢紊乱探讨益气调脂八味汤对CD40-CD40L系统的调控作用

目的:通过观察中药复方益气调脂八味汤对载脂蛋白E基因敲除(ApoE~(-/-))小鼠主动脉粥样硬化斑块病理形态和CD40-CD40L系统信号的影响,探讨益气调脂八味汤抗动脉粥样硬化的作用机制。方法:40只8周龄雄性ApoE~(-/-)小鼠予高脂饲料喂养建立实验性动脉粥样硬化小鼠模型,至第12周龄时,ApoE~(-/-)小鼠随机分为模型对照组、氯吡格雷组、中药高剂量组和低剂量组。取10只8周龄近交系C57BL/6小鼠作为正常对照组给予普通饲料喂养。各组于第24周取材检测血脂、外周血血小板表面PAC-1、CD40L表达;观察主动脉组织病理形态学改变;检测主动脉组织CD40L表达水平。结果:与模型对照组比较,中药2个剂量组小鼠血清TC、TAG和LDL-C水平均降低,HDL-C水平均升高,差异有统计学意义;与高剂量组比较,低剂量组小鼠血清TC、TAG、LDL-C和HDL-C水平比较差异均无统计学意义。中药组病变程度明显轻于模型对照组,并较氯吡格雷组轻。与模型对照组比较,中药个剂量组小鼠血清PAC-1、CD40L表达的百分比均明显降低,差异有统计学意义;与高剂量组比较,低剂量组小鼠血清PAC-1、CD40L表达百分比差异均无统计学意义。模型组动脉组织CD40L表达光密度值明显高于正常对照组;与高剂量组相比,低剂量组CD40L平均光密度值比较差异无统计学意义。结论:中药复方益气调脂八味汤能够调节血脂,调控CD40-CD40L系统的信号表达而抑制血小板活化,具有明显的抗动脉粥样硬化作用,且不存在剂量差异。 Objective: To observe the effect of Yiqi Tiaozhi Bawei Tang on aortic atherosclerotic plaque pathology and CD40-CD40L signaling in ApoE ~ (- / -) mice, To investigate the anti-atherosclerosis mechanism of Yiqi Tiaozhi Bawei Tang. Methods: Forty experimental Atherosclerotic mice were fed with high fat diet for 40 weeks old male ApoE ~ (- / -) mice. At week 12, ApoE ~ (- Divided into model control group, clopidogrel group, traditional Chinese medicine high-dose group and low-dose group. Ten eight-week-old inbred strains of C57BL / 6 mice were used as normal control group and fed with normal diet. At 24th week, the expression of PAC-1 and CD40L on the platelets of blood lipids and peripheral blood were detected. The pathological changes of the aorta were observed. The expression of CD40L in the aorta was detected. Results: Compared with the model control group, the levels of TC, TAG and LDL-C in the two groups of Chinese medicine were decreased and the levels of HDL-C were increased, the difference was statistically significant; compared with the high dose group, the low dose group There was no significant difference in serum TC, TAG, LDL-C and HDL-C levels in mice. The degree of lesion in the TCM group was significantly lower than that in the model control group and lighter than that in the clopidogrel group. Compared with the model control group, the percentages of the expression of PAC-1 and CD40L in the serum of the mice in each dose group were significantly decreased, the difference was statistically significant; compared with the high-dose group, the percentage of the expression of PAC-1 and CD40L The difference was not statistically significant. The optical density of CD40L in the model group was significantly higher than that in the normal control group. Compared with the high dose group, the average optical density of CD40L in the low dose group had no significant difference. Conclusion: The Chinese herbal compound Yiqi Tiaozhi Bawei Tang can regulate blood lipid, regulate the expression of CD40-CD40L system and inhibit platelet activation, and has obvious anti-atherosclerotic effect with no dose difference.
