在23例体外培养的人垂体GH分泌瘤细胞观察Ca~(2+)通道阻断剂尼卡地平和尼非地平、Ca~(2+)载体A23187对GH分泌瘤细胞GH基础分泌和GRH/SRIF类似物SMS刺激/抑制GH分泌的影响,同时观察GRH和SMS对~(45)Ca内流的作用。发现大多数GH瘤的GH分泌依赖Ca~(2+),但由Ca~(2+)介导的GH分泌过程有不同环节的异常,在GRH和SRIF调节Ca~(2+)系统中分别有66.7%(6/9例)和55.6%(5/9例)的瘤细胞培养存在受体及受体后Ca~(2+)通道和Ca~(2+)-GH分泌偶联异常,这些异常可能与GH瘤的高分泌有关。
The GH basal secretion of GH secreting tumor cells and GRH/SRIF were similarly observed in 23 human pituitary GH-secreting tumor cells cultured in vitro and Ca~(2+) channel blockers nicardipine and nifidipine, Ca~(2+) vector A23187. The SMS stimulated/inhibited the effects of GH secretion while observing the effects of GRH and SMS on ~(45)Ca influx. The GH secretion of most GH tumors was found to be dependent on Ca 2+ , but the GH secretion process mediated by Ca 2+ had abnormalities at different stages. There were 66% of GR 2+ and SRIF-regulated Ca 2+ systems, respectively. In 7% (6/9 cases) and 55.6% (5/9 cases) of tumor cell cultures, abnormal coupling of Ca~(2+) channels and Ca~(2+)-GH secretion was observed in the presence of receptors and receptors. Abnormalities may be related to the high secretion of GH tumors.
