目的初步表征分析大花红景天多糖的结构。方法采用乙醇分级沉淀和葡萄糖凝胶柱层析法分离纯化,得大花红景天多糖的主要成分(RCPS)。GC法进行一级结构分析的基础上,采用IR、UV、CD、SEM等手段对多糖的光谱学特性、不对称性以及形貌特征进行综合考察,同时还采用热分析仪分析热特性,结果与结论RCPS是一酸性杂多糖,相对重均分子量为2.7876×104,由鼠李糖、阿拉伯糖、木糖、甘露糖、葡萄糖、半乳糖和半乳糖醛酸组成,其摩尔比为1:2.96:0.21:0.26:0.08:0.58:0.15。RCPS在溶液中可能以无规则的方式存在,其粒度大约1~2μm,同时晶体间有非常微小的空隙。加热过程中,多糖出现了3次明显的质量损失。
Objective To characterize and analyze the structure of Rhodiola sachalinensis polysaccharide. Methods Separation and purification by ethanol fractionation and glucogel column chromatography were used to obtain the main component (RCPS) of Rhodiola crenulata polysaccharide. On the basis of primary structure analysis by GC method, IR, UV, CD, SEM and other methods were used to comprehensively examine the spectroscopic properties, asymmetry, and morphology of polysaccharides. Thermal characteristics were also analyzed using a thermal analyzer. And Conclusion RCPS is an acidic heteropolysaccharide with a relative weight average molecular weight of 2.7876×104 and is composed of rhamnose, arabinose, xylose, mannose, glucose, galactose and galacturonic acid in a molar ratio of 1:2.96. : 0.21:0.26:0.08:0.58:0.15. The RCPS may exist in a solution in an irregular manner, with a particle size of about 1 to 2 μm, and very small gaps between the crystals. During the heating process, there were three significant mass loss of polysaccharides.
