目的:探讨白藜芦醇(Res)对小鼠胚胎成纤维细胞(MEF)增殖和细胞周期的影响及变化规律。方法:通过不同浓度(1~150μmol/L)Res作用于增殖期的MEF,用MTT法检测72h后各组细胞增殖变化,用流式细胞术检测并分析其细胞周期。结果:Res1μmo/L组A值明显高于对照组(P<0.05),Res50μmol/L和150μmol/L组A值分别为0.196±0.055和0.185±0.034,均明显低于对照组(分别P<0.05,P<0.01),其细胞生长抑制率分别为14.0%和18.9%。细胞周期检测结果表明,Res1μmol/L组中G2/M期细胞占(14.82±0.49)%显著高于对照组(P<0.01)。50μmol/L和150μmol/L组G0/G1期细胞所占比例分别为(59.61±6.03)%和(65.31±0.27)%均显著高于对照组(P<0.01)。结论:Res对MEF增殖的影响,具有双向浓度依赖性,增殖和抑制增殖均与细胞周期调控有关,其中阻断G1期细胞进入S期是其抑制MEF增殖主要因素。
Objective: To investigate the effects of resveratrol on the proliferation and cell cycle of mouse embryonic fibroblasts (MEFs) and their changes. Methods: The proliferation of MEF was induced by resveratrol (1 ~ 150μmol / L) at different concentrations. The proliferation of MEF was detected by MTT assay after 72 hours. The cell cycle was detected by flow cytometry. Results: The A value in Res1μmo / L group was significantly higher than that in control group (P <0.05). The A values in Res50μmol / L and 150μmol / L group were 0.196 ± 0.055 and 0.185 ± 0.034, respectively, , P <0.01). The cell growth inhibition rates were 14.0% and 18.9%, respectively. Cell cycle analysis showed that cells in G2 / M phase in Res1μmol / L group (14.82 ± 0.49)% were significantly higher than those in control group (P <0.01). The proportion of cells in G0 / G1 phase in the groups of 50μmol / L and 150μmol / L were (59.61 ± 6.03)% and (65.31 ± 0.27)%, respectively, which were significantly higher than that of the control group (P <0.01). Conclusion: The effect of Res on proliferation of MEF cells in a bidirectional concentration-dependent manner. Proliferation and inhibition of proliferation are related to the regulation of cell cycle. Blocking G1 phase cells into S phase is the main factor that inhibits MEF proliferation.
