目的:观察补肾填精方对γ射线所致辐射损伤小鼠模型的作用,并通过观察其对DNA损伤修复ATM/ATR通路的影响,探讨该方抗辐射的作用机制。方法:将雌雄各半的balb/c小鼠,分成正常组、病理组和补肾组。采用γ射线损伤小鼠模型,在造模后取脾脏运用real-time RT-PCR方法检测该通路关键基因ATM和ATR基因的表达,并应用免疫组化方法检测ATM和ATR蛋白表达,结合图像分析技术进行分析。结果:补肾组ATR基因表达与病理组相比,有显著差异(P<0.05),而ATM基因表达在各组中的差异不明显;免疫组化结果与RT-PCR结果一致。结论:补肾填精方可促进ATR基因和蛋白的表达增强。
OBJECTIVE: To observe the effect of Bushen Tianjing Recipe on radiation-induced γ-induced radiation injury in mice and to observe its effect on DNA damage repair ATM / ATR pathway and to explore its mechanism. Methods: Male and female balb / c mice were divided into normal group, pathological group and Bushen group. The model of ATM and ATR was detected by real-time reverse transcription-polymerase chain reaction (RT-PCR). The expression of ATM and ATR genes was detected by immunohistochemistry. Combined with image analysis Technical analysis. Results: The expression of ATR gene in kidney transplantation group was significantly different from that in pathological group (P <0.05), while the expression of ATM gene in each group was not obvious. The results of immunohistochemistry were consistent with those of RT-PCR. Conclusion: Bushen Tianjing Recipe can promote the expression of ATR gene and protein.
