目的:探讨OPN反义基因(ANOPN)对人乳腺癌细胞系MDA-MB-231细胞生长及迁移能力的影响。方法:构建人骨桥蛋白反义基因真核表达质粒pcDNA3.1-ANOPN,将质粒pcDNA3.1-ANOPN、空载体pcDNA3.1(+)和等量的脂质体分别转染人乳腺癌细胞系MDA-MB-231,将转染细胞分别命名为MDA-ANOPN、MDA-vect和MDA,在体外观察各组细胞的迁移能力。结果:MDA-MB-231细胞比MDA-vect及MDA细胞迁移能力弱(P<0.01)。结论:ANOPN可抑制人乳腺癌细胞系MDA-MB-231细胞的迁移能力。
Objective: To investigate the effect of OPN antisense gene (ANOPN) on the growth and migration of human breast cancer cell line MDA-MB-231. Methods: The eukaryotic expression plasmid pcDNA3.1-ANOPN of human osteopontin antisense gene was constructed. The pcDNA3.1-ANOPN plasmid, empty vector pcDNA3.1 (+) and liposome were transfected into human breast cancer cell line MDA-MB-231, the transfected cells were named MDA-ANOPN, MDA-vect and MDA, in vitro observation of the migration of cells in each group. Results: The migration ability of MDA-MB-231 cells was lower than that of MDA-vect and MDA cells (P <0.01). Conclusion: ANOPN can inhibit the migration of human breast cancer cell line MDA-MB-231 cells.
