用RT-PCR及RACE方法从盐生植物大叶补血草Limonium gmelinii(Willd.)Kuntze中分离出Na+/H+逆向转运蛋白基因的cDNA(LgSOS1,GenBank登录号EU780458),LgSOS1的cDNA全长为3910bp,5′非翻译区为79bp,3′非翻译区为376bp,开放阅读框为3455bp,编码1151个氨基酸,推测分子量为126kD。氨基酸同源性分析表明,LgSOS1与冰叶午时花、沙拐枣、番茄、盐芥和拟南芥质膜型Na+/H+逆向转运蛋白的一致性分别为69.02%,64.42%,61.96%,60.12%和59.62%。预测分析表明,LgSOS1有12个跨膜结构区域。系统发育分析表明该蛋白与质膜型的Na+/H+逆向转运蛋白的亲缘关系较近,与液泡型的Na+/H+逆向转运蛋白亲缘关系较远。
The cDNA of Na + / H + antiporter gene (LgSOS1, GenBank accession number EU780458) was isolated from the halophyte Limonium gmelinii (Willd.) Kuntze by RT-PCR and RACE. The full-length cDNA of LgSOS1 was 3910 bp , The 5 ’untranslated region was 79 bp, the 3’ untranslated region was 376 bp, the open reading frame was 3455 bp, encoding a protein of 1151 amino acids, suggesting a molecular weight of 126 kD. Amino acid homology analysis showed that the identities of LgSOS1 and Na + / H + antiporter were 69.02%, 64.42%, 61.96% and 60.12 respectively % And 59.62%. Predictive analysis shows that LgSOS1 has 12 transmembrane structural regions. Phylogenetic analysis showed that this protein is closely related to the plasma membrane type Na + / H + antiporter and has a close relationship with the vacuolar Na + / H + antiporter.
