目的探讨基因与脑胶质瘤的相关性及其表达改变对胶质瘤U87细胞凋亡和增殖能力的影MACC1响。方法荧光定量PCR检测45例脑胶质瘤及相应癌旁组织中MACC1基因的表达;设计并合成基因特异MACC1性的siRNA,转染U87细胞;Western blot检测转染后MACC1蛋白的表达;双标流式细胞法检测细胞凋亡;MTT法测定细胞生长抑制率。结果基因在脑胶质瘤组织中表达明显上调;转染基因特异性的siRNA后MACC1蛋白表达明显降低,凋亡明显增加,生长抑制率明显增加,细胞增殖活力明显降低。结论 MACC1基因的表达异常与脑胶质瘤的发生相关,抑制其表达能促进肿瘤细胞的凋亡同时抑制细胞增殖。
Objective To investigate the relationship between gene and glioma and its effect on the apoptosis and proliferation of glioma U87 cells MACC1 ring. Methods The expression of MACC1 gene in 45 gliomas and corresponding paracancerous tissues was detected by real-time PCR. The gene-specific MACC1 siRNA was designed and synthesized and transfected into U87 cells. The expression of MACC1 protein was detected by Western blot. Cell apoptosis was detected by flow cytometry. Cell growth inhibition rate was measured by MTT assay. Results The gene expression was significantly up-regulated in glioma tissues. After transfection with gene-specific siRNA, the expression of MACC1 protein was significantly decreased, the apoptosis was significantly increased, the growth inhibition rate was significantly increased, and the cell proliferation activity was significantly decreased. Conclusion The abnormal expression of MACC1 gene is related to the occurrence of glioma. Inhibition of its expression can promote the apoptosis of tumor cells and inhibit the proliferation of tumor cells.
