目的 :探讨FHIT基因与胃肠肿瘤的关系。方法 :采用RT PCR及PCR产物直接测序法 ,检测了 2 6例胃癌组织和 30例大肠癌组织及其配对正常组织的FHIT基因。结果 :7例 (2 6 9% )胃癌组织和 8例 (2 6 7% )大肠癌组织存在异常FHIT转录本 ,而配对正常组织均无异常FHIT转录本 ;测序证实异常转录本缺失 1~ 3个FHIT外显子。结论 :异常FHIT转录本的表达是胃肠肿瘤发生中常见的和特有的改变 ;FHIT基因异常与胃肠肿瘤的发生和发展有关。
Objective: To investigate the relationship between FHIT gene and gastrointestinal tumors. Methods: The FHIT gene of 26 cases of gastric carcinoma and 30 cases of colorectal carcinoma and matched normal tissues were detected by RT-PCR and PCR products. RESULTS: FHIT transcripts were found in 7 cases (269%) of gastric cancer and 8 cases (267%) of colorectal cancers, but no abnormal FHIT transcripts were found in matched normal tissues. The deletion of abnormal transcripts was confirmed in 1 to 3 FHIT exons. Conclusion: The expression of abnormal FHIT transcripts is a common and specific change in the development of gastrointestinal tumors. The abnormality of FHIT gene is associated with the occurrence and development of gastrointestinal tumors.
