目的 观察牛磺酸对尾加压素Ⅱ (UrotensinⅡ ,UⅡ )抑制L 型钙电流的调节作用 ,并分析其作用机制及病理生理意义。方法 采用膜片钳全细胞记录方法 ,分别测定牛磺酸、UⅡ、牛磺酸 +UⅡ对豚鼠心室肌细胞L 型钙电流的影响。结果 在正常细胞外钙浓度时 ,(1)牛磺酸 5mmol L、10mmol L使L 型钙电流峰值分别下降 45 .1%、35 .4% ;(2 )UⅡ 10 - 9mol L使L 型钙电流峰值下降 6 9.4% (P <0 .0 5 ) ;但先给予牛磺酸 5mmol L、10mmol L后 ,再加入UⅡ 10 - 9mol L ,L 型钙通道电流峰值分别下降 5 3.4%、37.9% (P>0 .0 5 )。结论 牛磺酸可使UⅡ对L 型钙电流的抑制作用减弱 ,具有较强的钙调节作用及心肌保护作用。
Objective To observe the effect of taurine on the inhibition of L-type calcium currents by urotensin Ⅱ (UⅡ) and its mechanism of action and pathophysiological significance. Methods The patch clamp whole cell recording method was used to measure the effect of taurine, UⅡ, taurine and UⅡ on the L-type calcium current of guinea pig ventricular myocytes. Results In normal extracellular calcium concentration, (1) taurine 5 mmol L and 10 mmol L decreased the peak value of L - type calcium current by 45.1% and 35.4%, respectively; (2) U Ⅱ 10 - 9 mol L increased L - type calcium (P <0.05). However, the peak current of L-type calcium channel decreased by 5 3.4% and 37.9% after the addition of 10 mmol L of taurine and 10 mmol L of UII, (P> 0.05). Conclusion Taurine can reduce the inhibitory effect of UⅡ on L-type calcium current, and has strong calcium regulation and myocardial protection.
