目的:探讨小叶锦鸡儿黄酮类成分对实验性脑缺血再灌注损伤大鼠的作用。方法:以正常大鼠作为空白对照组,采用大脑中动脉阻断(MCAO)制备大鼠局灶性脑缺血再灌注模型,以阿司匹林(12.5 mg.kg-1)作为阳性对照,小叶锦鸡儿总黄酮(CMF)按照25,50,100 g.kg-1剂量,灌胃给药4周,观察给药后对MCAO大鼠脑组织超氧化物歧化酶(SOD)、丙二醛(MDA)含量的影响,并测定其全血黏度和血浆黏度(η),血小板聚集率(PAgT),红细胞聚集指数(EAI),红细胞刚性指数(IR),红细胞变形指数(DI)。结果:通过比较各组大鼠血流变学指标η(mPa.s),PAgT(%),EAI,IR,DI时发现,阿司匹林组[(1.71±0.08)mPa.s,(56.49±3.74)%,8.83±0.59,3.94±0.22,0.69±0.08)];CMF中剂量组[(1.72±0.11)mPa.s,(57.67±4.31)%,8.30±0.92,3.99±0.22,0.73±0.06)]、高剂量组(1.57±0.10)mPa.s,(55.59±4.58)%,7.83±0.58,3.65±0.17,0.70±0.07)较模型组大鼠[(1.96±0.09)mPa.s,(65.69±4.59)%,10.75±0.79,4.89±0.17,0.45±0.05)]的η,PAgT显著降低、红细胞参数EAI,IR显著降低,DI升高;通过比较各组大鼠脑组织SOD(U.mL-1)活性、MDA(nmol.mL-1)含量时发现,与模型组(50.21±10.74)U.mL-1,(1.61±1.07)nmol.mL-1相比,CMF低剂量组(55.45±11.67)U.mL-1,(1.38±0.96)nmol.mL-1、中剂量组(60.23±13.59)U.mL-1,(1.17±0.44)nmol.mL-1、高剂量组(64.58±9.36)U.mL-1,(1.08±0.42)nmol.mL-1均可提高MCAO大鼠脑组织SOD活性,降低MDA含量。结论:CMF对脑缺血再灌注损伤具有脑保护作用,这可能与其抗氧化应激、降低血黏度有关。
Objective: To investigate the effects of flavonoids from Caragana microphylla Lam on experimental cerebral ischemia-reperfusion injury in rats. Methods: The model of focal cerebral ischemia-reperfusion in rats was established by middle cerebral artery occlusion (MCAO) with normal rats as blank control group. Aspirin (12.5 mg.kg-1) was used as positive control and Caragana microphylla Total flavonoids (CMF) were administered orally for 25 weeks at doses of 25, 50, 100 g.kg-1 for 4 weeks. The levels of superoxide dismutase (SOD), malondialdehyde (MDA) The whole blood viscosity and plasma viscosity (η), platelet aggregation rate (PAgT), erythrocyte aggregation index (EAI), erythrocyte rigidity index (RI) and erythrocyte deformability index (DI) were measured. Results: Compared to the hemorheological parameters η (mPa.s), PAgT (%), EAI, IR and DI, the aspirin group [(1.71 ± 0.08) mPa.s, (56.49 ± 3.74) (1.72 ± 0.11) mPa.s, (57.67 ± 4.31)%, 8.30 ± 0.92, 3.99 ± 0.22, 0.73 ± 0.06)]; (1.97 ± 0.10) mPa.s, (55.59 ± 4.58)%, 7.83 ± 0.58,3.65 ± 0.17,0.70 ± 0.07 in high dose group compared with [(1.96 ± 0.09) mPa.s, (65.69 ± 4.59)%, 10.75 ± 0.79,4.89 ± 0.17,0.45 ± 0.05)] η, PAgT decreased significantly, erythrocyte parameters EAI, IR decreased significantly, DI increased; By comparing the brain tissue SOD (U.mL- 1) activity and the content of MDA (nmol.mL-1) in the CMF low-dose group (55.45 ± 0.51) compared with the model group (50.21 ± 10.74) U.mL- (1.17 ± 0.44) nmol.mL-1, and high-dose group (64.58 ± 11.67) U.mL-1, 1.38 ± 0.96 nmol.mL-1 and 60.23 ± 13.59 U.mL- 9.36) U.mL-1, (1.08 ± 0.42) nmol.mL-1 can increase the activity of SOD in brain tissue of MCAO rats and decrease the content of MDA. Conclusion: CMF has a protective effect on cerebral ischemia-reperfusion injury, which may be related to its anti-oxidative stress and lower blood viscosity.
