目的观察六味地黄丸对TNF-α、IL-6表达水平的影响及糖尿病肾病大鼠肾脏的保护机制的研究。方法复制DN大鼠模型,将其分为正常组、模型组、治疗组(六味地黄丸高、中、低剂量组及罗格列酮组),全自动生化分析仪测大鼠肾功能、考马斯亮蓝法测24 h尿蛋白量情况,Elisa法检测血清中TNF-α、IL-6的水平,同时用葡萄糖氧化酶法测定空腹血糖(FBG)。结果六味地黄丸高、中剂量组及罗格列酮组大鼠24 h尿蛋白量、血Scr、BUN均低于模型组(P<0.01),且高剂量组疗效与与罗格列酮组差异无统计学意义(P>0.05);同时各治疗组TNF-α表达水平较模型组下调(P<0.01),其中六味地黄丸高剂量组与罗格列酮组之间差异无统计学意义(P>0.05);而六味地黄丸高、中剂量组和罗格列酮组IL-6在肾脏的表达均高于模型组(P<0.01),且高剂量组和罗格列酮缬组在表达上差异无统计学意义(P>0.05)。结论六味地黄丸可用于治疗早期糖尿病肾病,对肾脏有保护作用,机制可能与其能抑制DN大鼠肾脏中TNF-α、IL-6的激活有关。
Objective To observe the effect of Liuweidihuangwan on the expression of TNF-α and IL-6 and the protective mechanism of kidney in diabetic nephropathy rats. Methods DN model rats were duplicated and divided into normal group, model group, treatment group (high, medium and low dose of Liuweidihuangwan group and rosiglitazone group) and renal function of rats by automatic biochemical analyzer Serum Brilliant Blue assay 24 h urinary protein content, Elisa method detection of serum levels of TNF-α, IL-6, while using glucose oxidase method for the determination of fasting blood glucose (FBG). Results The urinary albumin, Scr and BUN in high and middle dose Liuwei Dihuang Wan group and rosiglitazone group were lower than those in model group (P <0.01), and the effect of high dose group and rosiglitazone group The difference was not statistically significant (P> 0.05). At the same time, the expression of TNF-α in each treatment group was lower than that in model group (P <0.01), and there was no significant difference between high-dose Liuweidihuangwan group and rosiglitazone group (P> 0.05). The expression of IL-6 in high, medium and rosiglitazone groups of Liuweidihuangwan group was higher than that of model group (P <0.01), and the high dose group and rosiglitazone group There was no significant difference in expression (P> 0.05). Conclusion Liuweidihuangwan can be used to treat diabetic nephropathy in early stage and protect the kidney. The mechanism may be related to the inhibition of TNF-α and IL-6 in the kidney of DN rats.
