建立UPLC-PDA(超高效液相色谱-二极管阵列检测器)同时测定不同产地雪菊和市售菊花样品中4种多酚类化合物(儿茶素、3,5-o-二咖啡酰基奎宁酸、木犀草苷、槲皮素)方法。色谱条件:JADE-PAK CB-C18液相色谱柱(100 mm×2.1 mm,3μm),流动相为甲醇-0.05%甲酸溶液,进样量1μL,梯度洗脱,流速0.2 mL/min,柱温25℃,检测波长采用双通道(278 nm和350 nm)。结果表明,各组分的质量浓度与峰面积具有良好的线性关系,相关系数均大于0.999,各组分在20 min内得到了较好的分离。方法定量限0.10~0.95 g/kg,平均回收率高于91.1%,相对标准偏差低于4.2%。与市售菊花相比,雪菊在多酚类化合物总含量具有一定的优势。方法预处理简单,准确度高且分析时间短,可用于不同产地雪菊和市售菊花样品中多酚类化合物的快速质量评价。
UPLC-PDA (Ultra Performance Liquid Chromatography-Diode Array Detector) was developed for the simultaneous determination of four polyphenols (catechin, 3,5-o-dicoferate quinine Acid, luteolin, quercetin) method. Chromatographic conditions: JADE-PAK CB-C18 column (100 mm × 2.1 mm, 3 μm) with a mobile phase of methanol-0.05% formic acid and injection volume of 1 μL with gradient elution at a flow rate of 0.2 mL / 25 ℃, detection wavelength dual-channel (278 nm and 350 nm). The results showed that the mass concentration of each component had a good linear relationship with the peak area, the correlation coefficient was greater than 0.999, and the components were well separated within 20 min. The limits of quantification were 0.10 ~ 0.95 g / kg, the average recovery was higher than 91.1% and the relative standard deviation was less than 4.2%. Compared with commercially available chrysanthemum, snow chrysanthemum in polyphenols total content has certain advantages. The method has the advantages of simple pretreatment, high accuracy and short analysis time, and can be used for rapid quality evaluation of polyphenols in snowdrops and chrysanthemum samples from different areas.
