                                目的 :探索 p16基因和 p5 3基因及 p16基因和顺铂(CDDP)联合应用对体内外胆管癌细胞系生长的抑制作用 .方法 将重组体腺病毒 p16 (Ad- p16 )和 Ad- p5 3及 Ad- p16和CDDP联合作用于人胆管癌细胞系 QBC939,观察和分析其对体内外胆管癌细胞生长抑制作用及其机制 .结果  Ad- p16在 QBC939细胞中表达能抑制 QBC939细胞的生长 ,与 p5 3、CDDP联合应用能明显抑制 QBC939细胞的生长 ,p16和 p5 3基因诱导癌细胞发生 G1期阻滞和细胞凋亡 ,CDDP能诱导癌细胞发生 G2期阻滞和细胞凋亡 ;裸鼠皮下移植瘤模型瘤体内注射 Ad- p16及腹腔内注射 CDDP均能抑制 QBC939肿瘤的生长 ,两者联合应用具有协同作用 .结论  p16基因和 p5 3基因及 CDDP联合应用具有协同作用 .
OBJECTIVE: To investigate the inhibitory effect of the combination of p16 gene, p5 3 gene, p16 gene and cisplatin (CDDP) on the growth of human cholangiocarcinoma cell lines in vitro and in vivo.Methods The recombinant adenovirus p16 (Ad-p16) and Ad-p5 3 Ad-p16 and CDDP combined with human cholangiocarcinoma cell line QBC939 to observe and analyze its inhibitory effect on the growth of human cholangiocarcinoma cells and its mechanism.Results Ad-p16 expression in QBC939 cells can inhibit the growth of QBC939 cells, and p5 CDDP can significantly inhibit the growth of QBC939 cells, p16 and p5 3 gene induced G1 arrest and apoptosis in cancer cells, CDDP can induce G2 arrest and apoptosis in cancer cells; nude mice subcutaneous transplantation Tumor model in vivo injection of Ad-p16 and intraperitoneal injection of CDDP can inhibit the growth of QBC939 tumor, the combination of the two have synergistic effect.Conclusion The combination of p16 gene and p5 3 gene and CDDP has a synergistic effect.
