目的:探讨基质金属蛋白酶-2(MMP-2)与妊娠期高血压疾病子痫前期(PE)发病的关系。方法:采用明胶酶谱法检测14例正常晚孕妇女和38例PE孕妇母血和脐血血清中MMP-2及其酶原pro-MMP-2的水平。结果:PE组及对照组的母血及脐血中均有MMP-2及pro-MMP-2的表达。PE组母血中MMP-2水平显著升高(P<0.05),并随病情加重而增加。PE组母血pro-MMP-2、脐血中MMP-2及pro-MMP-2均无显著改变(P>0.05)。新生儿的结局与孕母血清中MMP-2水平具有一定相关性。结论:PE患者妊娠晚期MMP-2与滋养层侵袭能力下降关系不大,而PE母血中MMP-2的升高可能与血管内皮功能紊乱有关。MMP-2有望成为预测妊娠期高血压疾病严重程度及新生儿结局的一项生化指标。
Objective: To investigate the relationship between matrix metalloproteinase-2 (MMP-2) and preeclampsia (PE) in pregnancy-induced hypertension. Methods: The levels of MMP-2 and proenzyme MMP-2 in maternal serum and umbilical cord blood of 14 normal pregnant women and 38 pregnant women were detected by gelatin zymography. Results: The expression of MMP-2 and pro-MMP-2 in maternal blood and umbilical cord blood of PE group and control group respectively. The level of MMP-2 in maternal blood of PE group increased significantly (P <0.05), and increased with the aggravation of the disease. There was no significant change in pro-MMP-2 and pro-MMP-2 in maternal blood of PE group (P> 0.05). Neonatal outcome and maternal serum MMP-2 levels have a certain relevance. Conclusions: There is little correlation between MMP-2 and invasiveness of PE in pregnant women with PE. However, the increase of MMP-2 in PE maternal blood may be related to endothelial dysfunction. MMP-2 is expected to be a biochemical marker for predicting the severity of neonatal hypertensive disorders in pregnancy.
