目的 :了解 WT1基因在各造血系统肿瘤中的表达。方法 :应用筑巢式 RT- PCR检测 63例血液系肿瘤 WT1基因表达。结果 :急性白血病初诊时 WT1基因的表达率 81.9% ,完全缓解期的表达率 60 .0 % ;慢性白血病慢性期或加速期并不表达 WT1基因 ,但慢粒进展至原始细胞危象期 ,均表达 WT1基因。造血系统的其它恶性病 ,如恶性组织细胞病和骨髓增生异常综合征的表达率分别为60 .0 %和 58.3%。正常人外周血并不表达 WT1基因 ,造血系统非肿瘤性疾病的骨髓有核细胞 WT1基因表达率极低 ( 1/ 10 )。结论 :WT1基因的异常高表达可能与造血系统的恶性肿瘤发病有关。WT1基因的 RT- PCR检测可作用白血病残留细胞病检测的一个有用的辅助指标。
Objective : To understand the expression of WT1 gene in hematopoietic system tumors. Methods : Nested RT-PCR was used to detect the expression of WT1 gene in 63 blood tumors. Results: The expression rate of WT1 gene was 81.9% in the initial diagnosis of acute leukemia and 60.0% in the complete remission period. WT1 gene was not expressed in the chronic or accelerated phase of chronic leukemia, but the slow-granulocyte progressed to the protocell crisis phase. Expression of WT1 gene. The other malignant diseases of the hematopoietic system, such as malignant histiocytosis and myelodysplastic syndrome, were respectively 60.0 % and 58.3 %. Normal human peripheral blood does not express the WT1 gene, and the WT1 gene expression rate of bone marrow nucleated cells in hematopoietic system non-neoplastic diseases is extremely low (1/10). Conclusion: The abnormally high expression of WT1 gene may be related to the pathogenesis of malignant tumors in the hematopoietic system. The RT-PCR detection of the WT1 gene can be used as a useful auxiliary marker for the detection of leukemia residual cell disease.
