目的:观察肾复宁颗粒对慢性肾功能衰竭大鼠的作用及可能机制。方法:SD大鼠采用右肾切除左肾外动脉分支结扎方法建立CRF模型,随机分为模型对照组、雷公藤多甙片组(0.006g/kg)、肾复宁颗粒组(9g/kg)、肾复宁颗粒组(6g/kg)、肾复宁颗粒组(3g/kg),各15只,另设假手术组15只,连续给药8周,观察血清Scr、BUN、Cys-C含量及肾脏组织中TGF-β1mRNA、细胞凋亡基因Fas/Fas L表达水平。结果:治疗后,治疗组及雷公藤多甙片组大鼠血清中Scr、BUN、Cys-C水平明显下降,大鼠肾脏组织中TGF-β1 mRNA、Fas/Fas L表达减少,尤以肾复宁颗粒(9g/kg)显著。结论:肾复宁颗粒通过抑制慢性肾功能衰竭大鼠肾脏TGF-β1mRNA、Fas/Fas L表达,调控肾实质细胞调亡,从而改善肾功能,达到延缓肾脏纤维化作用,其疗效具有一定剂量依赖性。
Objective: To observe the effect and the possible mechanism of “Shenfeining Granules” in rats with chronic renal failure. Methods: The CRF model was established in SD rats by right nephrectomy and branchage of the left renal artery. The rats were randomly divided into three groups: model control group, Tripterygium wilfordii tablet group (0.006g / kg), Shenfuning granule group (9g / kg) , Shenfuning granule group (6g / kg) and Shenfining granule group (3g / kg), 15 rats in each group, and 15 rats in sham operation group for 8 weeks. Serum Scr, BUN, Cys-C Content and the expression of TGF-β1mRNA and Fas / Fas Ligand in renal tissue. Results: After treatment, the levels of Scr, BUN and Cys-C in the treatment group and the tripterygium glycosides tablet group decreased significantly, while the expression of TGF-β1 mRNA and Fas / Fas L decreased in the kidney tissue, especially in the kidney Ning particles (9g / kg) significant. Conclusion: Shenfuning granule can improve the renal function and delay the renal fibrosis by inhibiting the expression of TGF-β1mRNA, Fas / Fas L in the kidney of rats with chronic renal failure and regulating the apoptosis of renal parenchymal cells in a dose-dependent manner Sex.
