以玻璃微电极细胞内记录技术,研究了培养心肌细胞的自发电活动,测量了22个培养心肌细胞动作电位的正常参数。在培养液中加入氟累积达1.25ppm,2.5ppm,3.75ppm 和5ppm 时,动作电位各参数随氟剂量增加而发生以下变化:APA,OS 和 RP 逐渐降低,APD 缩短,Vmax 和 DDR 减慢,BR 减小,以上结果提示氟能影响钙内流和舒张期去极化电流而改变培养心肌细胞的电活动。
The intracellular recording technique of glass microelectrode was used to study the self-generating activity of cultured cardiomyocytes, and the normal parameters of action potentials of 22 cultured cardiomyocytes were measured. The parameters of action potential with the increase of fluoride dosage were as follows: APA, OS and RP gradually decreased, APD shortened, Vmax and DDR slowed down, while the addition of fluorine reached 1.25ppm, 2.5ppm, 3.75ppm and 5ppm. BR decreased, the above results suggest that fluorine can affect calcium influx and diastolic depolarization current and change the electrical activity of cultured cardiomyocytes.
