豚鼠离体心室肌经95%O2的甚氧液道流后,用5%、10%和20%O2的低氧值灌流,用微电极记录动作电位(AP),观察丹圣(H303)对缺氧引起的心室肌电活动变化的保护作用.结果表明,丹圣对缺O2前后心室肌细胞静息电位水平的变化无明显影响(P>0.05),但可保护缺O2引起的动作电位相值和动作电位时程的变化,后者尤为明显(P<0.001),随着缺O2持续时间的延长,丹圣对APD的作用减弱。说明丹圣对缺O2引起的心室肌细胞APA和APD的变化有保护作用。
After guinea pig isolated ventricle myocardium was subjected to a 95% O2 flow of a very oxygenic stream, hypoperfusion with 5%, 10%, and 20% O2 was perfused. The action potential (AP) was recorded with a microelectrode to observe the effect of Dansheng (H303) on Protective effects of changes in ventricular myoelectric activity caused by hypoxia. The results showed that Dansheng had no significant effect on the resting potential level of ventricular myocytes before and after O2 deficiency (P>0.05), but it could protect the change of action potential phase and action potential duration caused by O2 deficiency. This was especially significant (P<0.001). With the extension of the duration of O2 deficiency, Dansheng’s effect on APD weakened. It shows that Dansheng has a protective effect on the changes of APA and APD in ventricular myocytes induced by oxygen deficient.
