目的 观察骨肉瘤组织 、 和 型胶原蛋白和 m RNA的表达及其与骨肉瘤分型和分化的关系。方法 免疫组化 LSAB法检测胶原蛋白 ,原位杂交检测胶原 m RNA的表达 ,用图像分析系统进行分析和半定量测定。结果 骨肉瘤组织不仅表达 型胶原蛋白 (阳性率 87% ,灰度值1 71 .99± 1 4.74) ,并出现了 型和 型胶原蛋白 (阳性率分别为 30 .4%和 43.4% ,灰度值分别为1 5 3.0 7± 1 8.82和 1 68.2 9± 1 8.36) ;软骨母细胞型和混合型骨肉瘤 型胶原的表达高于其它型 ,差异有显著性 ( P<0 .0 1 ) ;高中低分化的三组骨肉瘤间 , 型胶原表达差异有显著性 ( P<0 .0 1 ) ,分化越低表达越少。 、 型胶原 m RNA表达与蛋白表达所得结论基本一致。结论 型胶原可作为软骨母细胞型和混合型骨肉瘤的分型参考指标 , 型胶原可作为骨肉瘤分化及恶性度判断的参考指标
Objective To observe the expression of collagen and m RNA in osteosarcoma and its relationship with osteosarcoma typing and differentiation. Methods Immunohistochemistry was used to detect collagen by LSAB method. In situ hybridization was used to detect the expression of collagen m RNA. The image analysis system was used for analysis and semi-quantitative determination. Results The osteosarcoma tissues expressed not only collagen (positive rate 87%, gray value 1 71 .99±1 4.74), but also type and type collagen (positive rates were 30.4% and 43.4%, respectively. The values were 1 5 3.0 7± 18.82 and 1 68.2 9± 1 8.36). The expression of chondroblastoma and mixed osteosarcoma collagen was higher than that of other types. The difference was significant (P < 0.01). Among the three groups of osteosarcoma with high-grade and low-differentiation, the expression of collagen was significantly different (P 0.01), and the lower the differentiation, the lower the expression. The results of mRNA expression and protein expression of type-collagen were basically consistent. Conclusion Collagen can be used as a reference for the classification of chondroblastoma and mixed osteosarcoma. Collagen can be used as a reference marker for differentiation and malignancy of osteosarcoma.
