目的:评价白细胞介素IL-12mRNA的表达与角膜移植排斥的关系。方法:采用反转录聚合酶链反应(RT-PCR)的方法定量检测大鼠同种异体角膜移植物中IL-2、12mRNA,并对角膜植片进行临床观察,以混浊、水肿和新生血管3项指标积分总和作为排斥反应指数(RI)进行临床评估。结果:同种异体组与对照组大鼠角膜植片在移植术后第12天RI分别为5.80±1.07和3.20±0.84,P=0.00003,同种异体组角膜植片内有大量的细胞介素2、12mRNA的表达。结论:IL-12mRNA的表达在同种异体角膜移植捧斥反应中有重要作用。
Objective: To evaluate the relationship between the expression of IL-12 mRNA and the rejection of corneal allografts. Methods: The levels of IL-2 and 12 mRNA in rat corneal allografts were quantitatively determined by reverse transcription-polymerase chain reaction (RT-PCR). The corneal graft was observed clinically. The turbidity, edema and neovascularization The sum of the three indicator scores was used as a clinical evaluation of rejection index (RI). Results: On the 12th day after transplantation, RI of allograft group and control group was 5.80 ± 1.07 and 3.20 ± 0.84, respectively, P = 0.00003. There were a large number of intracellular corneal allografts 2,12 mRNA expression. Conclusion: The expression of IL-12 mRNA plays an important role in the rejection of corneal allograft.
