目的:观察转化生长因子B1(Transforming gowth factor-β11TGF-β1)在阿霉素肾病(Doxorubicin.induced nephropathy DIN)大鼠疾病早期的表达及辛伐他汀对TGF-β1表达的影响。方法:将100只6周龄雄性sD大鼠随机分为正常组、肾病组和辛伐他汀治疗组3组,第7、21、42天检测各组大鼠胆固醇、甘油三酯,24小时尿蛋白定量、尿TGF-β1以及肾组织中TGF-β1。结果:(1)、21天辛伐他汀组与肾病组比较各项检查均无显著差异(P>0.05);第42天辛伐他汀组尿蛋白低于肾病组(P<0.05),有显著差异;尿TGF-β1、肾组织TGF-β1、胆固醇、甘油三酯有非常显著差异(P<0.01);(2)、肾病组肾组织TGF-β1与尿蛋白、尿TGF-β1成正相关。结论:TGF-β1与MCNS尿蛋白的发生有关;辛伐他汀可降低血脂及尿蛋白,降低TGF-β1的表达,减轻肾脏损害,延缓疾病进展。
Objective: To observe the expression of transforming growth factor-β1 (TGF-β1) in the early stage of Doxorubicin-induced nephropathy (DIN) and the effect of simvastatin on the expression of TGF-β1. Methods: 100 male SD rats of 6 weeks old were randomly divided into three groups: normal group, nephrosis group and simvastatin treatment group. Cholesterol, triglyceride and 24-hour urine of rats in each group were detected on days 7, 21 and 42 Protein quantitation, urinary TGF-β1, and TGF-β1 in renal tissue. Results: There was no significant difference between the simvastatin group and nephropathy group on the 21th day (P> 0.05); the proteinuria of the 42nd simvastatin group was lower than that of the nephropathy group (P <0.05) (P <0.01). (2) TGF-β1 in renal tissue was positively correlated with urinary protein and urinary TGF-β1. There was a significant difference between the two groups in urinary TGF-β1, TGF-β1, cholesterol and triglyceride. Conclusion: TGF-β1 is associated with the occurrence of urinary protein in MCNS. Simvastatin can reduce blood lipids and urinary protein, reduce the expression of TGF-β1, reduce renal damage and delay disease progression.
