目的 观察不同免疫缺陷小鼠中的人肝癌细胞生长情况以及 T,B淋巴细胞的免疫作用 ,探讨免疫缺陷小鼠肿瘤模型制作的意义 .方法 将体外培养的人肝癌细胞接种到四种免疫缺陷小鼠 ,分别为 :B细胞缺陷的 CBA/N,T细胞缺陷的 BAL B/c- nu,T,B细胞缺陷的 SCID及免疫重建的 SCID小鼠 ,观察其生长特点 ;作鼠脾细胞毒试验 ,测定外周血 CD4 + ,CD8+ 数分数和荷瘤鼠血清 Ig含量的变化 ;作肝癌细胞凝集试验 .结果 CBA/N和用 BAL B/c鼠外周血淋巴细胞重建的 SCID(B- PBL- SCID)鼠不成瘤 ,nude,SCID和用 CBA/N鼠外周血淋巴细胞重建的 SCID (C- PBL - SCID)小鼠全部成瘤 ;SCID鼠的瘤体比裸鼠瘤体长的更快 ,肝内接种转移率更高、转移范围更大 .接种瘤细胞的 BAL B/c和 CBA/N鼠脾细胞对癌细胞杀伤力较强 ,免疫重建的 SCID鼠脾细胞毒杀伤较小 .接种瘤细胞的鼠 CD4 +数分数都下降 ,CD8+变化不大 ,CD4 + /CD8+比值下降 .具有 B细胞的实验鼠均测得 Ig在 mg· L- 1 水平 ,并能使癌细胞产生凝集反应 .结论 SCID鼠是建立肿瘤模型和免疫重建研究的理想小鼠 ;小鼠成瘤率与 T细胞相关 ,T细胞在异种瘤移植排斥中起主要作用 ;B淋巴细胞及其产生的抗体在抗肿瘤中起着不可忽视的作用
