探讨热休克蛋白 70 (Hsp70 ) - H2 2肽复合物与小鼠骨髓来源的树突状细胞 (dendritic cell,DC)成熟之间的关系。采用 GM- CSF、IL- 4刺激培养小鼠骨髓细胞 ,增殖产生大量 DCs;从 H2 2肝癌细胞中获取肿瘤细胞肽 ,体外与Hsp70进行结合 ,结合产物修饰 DC,通过细胞因子检测试剂盒和流式细胞仪对修饰的 DCs上清液和 DCs膜表面分子进行检测。发现 Hsp70 - H2 2肽复合物修饰的 DCs大量分泌 IL- 12、TNF- α、IL- 1β等细胞因子 ,并且高表达主要组织相容性 类抗原 (MHC 类 )、 CD80、 CD40等分子 ,而单独的 Hsp70或 H2 2肽则不具有此作用。结果提示 :Hsp70 - H2 2肽复合物可诱导 DC成熟 ,这种成熟的 DC分泌的因子和表达的分子对于抗肿瘤免疫是非常有意义的
To investigate the relationship between heat shock protein 70 (Hsp70) - H2 2 peptide complex and mouse bone marrow-derived dendritic cell (DC) maturation. A large number of DCs were proliferated by using GM-CSF and IL-4 to stimulate the proliferation of mouse bone marrow cells. Tumor cell peptides were obtained from H 2 2 hepatoma cells and bound with Hsp70 in vitro. The DCs were modified with the product and detected by cytokine kit and flow cytometry Cytometry modified DCs supernatant and DCs membrane surface molecules were detected. It was found that DCs modified by Hsp70 - H2 2 peptide complex secreted a large number of cytokines such as IL - 12, TNF - α, IL - 1β and overexpressed major histocompatibility antigens (MHC class), CD80 and CD40 Hsp70 or H2 2 peptides alone do not have this effect. The results suggest that the Hsp70 - H2 2 peptide complex can induce DC maturation. The mature DC secreting factor and the expressed molecule are of great significance for antitumor immunity
