目的依据微生态学原理,针对紫藤蚜的肠道细菌进行分离鉴定,为探索紫藤蚜的营养生理与肠道细菌之间的关系等提供理论依据。方法利用传统的分离培养方法对紫藤蚜5株肠道细菌的生理生化性状进行系统研究,并针对细菌16S rDNA保守区设计通用引物进行PCR扩增、测序,将获得的16S rDNA序列与美国NCBI信息中心数据库中的发表序列细菌进行比对,分析克隆群中细菌的种属。结果分离纯化得到的5株细菌经过鉴定,其中1、2、3号菌属于蜡状芽孢杆菌属(Bacillus cereus)、4号菌为沙雷菌属(Serratia bzio)、5号菌为枯草芽孢杆菌属(Bacillus subtilis)。结论本研究为进一步探讨紫藤蚜生长发育与肠道细菌的关系以及深入研究菌株提供工作基础。
Objective To isolate and identify the intestinal bacteria of the wisteria aphid according to the principle of microecology and to provide the theoretical basis for exploring the relationship between the nutrition physiology of the wisteria aphid and the intestinal bacteria. Methods The physiological and biochemical characteristics of 5 strains of enterobacter aphids were systematically studied by the traditional method of isolation and culture. The universal 16S rDNA sequences were designed for PCR amplification and sequencing. The 16S rDNA sequences obtained were compared with the NCBI The published sequence bacteria in the central database were aligned to analyze the bacterial species in the clonal population. Results Five strains of bacteria isolated and purified were identified. Among them, No. 1, No. 2 and No. 3 bacteria belonged to Bacillus cereus, No. 4 strain was Serratia bzio, No. 5 strain was Bacillus subtilis Bacillus subtilis. Conclusion This study provides the basis of further study on the relationship between growth and development of gynogenetic aphids and gut bacteria and in-depth study of strains.
