目的:检测并验证新基因MTB139在儿童结核临床分离株中的表达,通过比较其在不同耐药性结核结核分枝杆菌中的表达差异,探讨其与结核耐药性的关系。方法:应用逆转录聚合酶链式反应(reverse transcription polymerase chain reaction,RT-PCR)的方法检测60例临床结核分离株(9例耐多药株,24例非耐多药株,27例敏感株)中基因MTB139的表达。结果:60例结核分离株样本中有7例检测到MTB139基因的阳性表达,3个组(耐多药组、非耐多药组、敏感组)结核菌株中MTB139基因阳性表达率比较有统计学差异(P=0.000),其中耐多药组MTB139基因阳性表达率(5例,阳性表达率55.56%)明显高于非耐多药组(1例,阳性表达率4.17%)及敏感组(1例,阳性表达率3.70%),差异有统计学意义(P=0.003、P=0.002),非耐多药组与敏感组MTB139基因阳性表达率无统计学差异(P=1.000)。结论:新基因MTB139在耐多药结核菌株有高表达,可能与结核耐多药性相关。
Objective: To detect and verify the expression of a new gene MTB139 in clinical isolates of children with tuberculosis, and to explore the relationship between the expression of MTB139 and the drug resistance of Mycobacterium tuberculosis by comparing the expression of MTB139 in different drug resistant tuberculosis. Methods: Sixty clinical TB isolates were detected by reverse transcription polymerase chain reaction (RT-PCR), including 9 multi-drug resistant strains, 24 non-multi-drug resistant strains and 27 sensitive ones ) Gene MTB139 expression. Results: The positive expression of MTB139 gene was detected in 7 out of 60 samples of tuberculosis isolates. The positive rates of MTB139 gene expression in three groups (multidrug-resistant group, non-multi-drug resistant group, sensitive group) were statistically significant (P = 0.000). The positive rate of MTB139 gene in MDR-resistant group (5 cases, the positive expression rate was 55.56%) was significantly higher than that in non-MDR group (1 case, positive rate 4.17% (P = 0.003, P = 0.002). There was no significant difference in the positive expression rate of MTB139 between non-resistant group and sensitive group (P = 1.000). Conclusion: The new gene MTB139 is highly expressed in MDR-TB strains, which may be related to the multi-drug resistance of tuberculosis.
