目的 研究去lL 6饥饿引发的人骨髓瘤细胞系XG 7凋亡以及三种Bcl 2家族抗凋亡蛋白 (Bcl 2 ,Bcl xL,Mcl 1)在凋亡发生过程中的作用。方法 采用碘化丙腚 (PI)染色和流式细胞分析术确定去IL 6饥饿引发的XG 7细胞凋亡效应 ;采用免疫印迹方法检测Bcl 2家族分子在XG 7细胞中的表达情况。结果 经去IL 6饥饿处理一定时间后 ,可观察到一定比例的凋亡XG 7细胞 ,同时三种抗凋亡蛋白之一的Mcl 1在XG 7细胞中的表达明显下调。在经过去因子饥饿处理的XG 7细胞中重新补加IL 6 ,则细胞凋亡被明显抑制 ,同时Mcl 1表达上调结论 Mcl 1在去IL 6饥饿所引发的XG 7细胞凋亡过程中具有重要作用 ,而Bcl 2 ,Bcl xL 不参与此凋亡反应
Objective To investigate the apoptosis of human myeloma cell line XG 7 induced by starvation and the roles of three Bcl 2 family anti-apoptotic proteins (Bcl 2, Bcl xL, Mcl 1) during apoptosis. Methods The apoptosis of XG 7 cells induced by IL-6 starvation was determined by propidium iodide (PI) staining and flow cytometry. The expression of Bcl-2 family members in XG 7 cells was detected by Western blotting. Results A certain percentage of apoptotic XG 7 cells were observed after being treated with IL-6 for a long time. Mcl 1, one of the three anti-apoptotic proteins, was significantly down-regulated in XG 7 cells. After reintroduction of IL-6 into factor-starved XG 7 cells, apoptosis was significantly inhibited and Mcl 1 was up-regulated. Conclusions Mcl 1 is important in the process of apoptosis of XG 7 cells induced by IL 6 starvation Bcl 2, Bcl xL are not involved in this apoptotic response
