目的:建立葛花中染料木苷和鸢尾苷两个异黄酮成分的HPLC含量测定方法。方法:Agilent ZORBAX Eclipse XDBC18色谱柱(4.6 mm×150 mm,5μm);以不同浓度的乙腈-1‰磷酸水为流动相梯度洗脱,流速0.8 ml·min-1,检测波长270nm,柱温30℃。结果:染料木苷和鸢尾苷的线性范围分别为46.7~747.2μg·ml-1(r=0.999 9)和54.1~864.8μg·ml-1(r=0.999 9);平均回收率分别为101.1%(RSD为1.46%)和100.2%(RSD为1.58%)。结论:本方法简单,专属性强,重复性好,可用于葛花药材的质量控制。
Objective: To establish a HPLC method for the determination of genistin and tectorigenin isoflavones in Pueraria lobata. METHODS: Agilent ZORBAX Eclipse XDBC18 column (4.6 mm × 150 mm, 5 μm) was used. The mobile phase consisted of acetonitrile-1 ‰ phosphoric acid with different concentrations as the mobile phase. The flow rate was 0.8 ml · min-1. ℃. Results: The linear range of genistin and irisoside were 46.7-747.2μg · ml-1 (r = 0.999 9) and 54.1-864.8μg · ml-1 (r = 0.999 9) respectively. The average recoveries were 101.1% (1.46% RSD) and 100.2% (RSD 1.58%). Conclusion: The method is simple, specific and reproducible, and can be used for the quality control of Gehua medicinal materials.
