目的:通过采用免疫亲和柱-高效液相色谱法来测定花生中的黄曲霉毒素。方法:样品用甲醇+水(70∶30)提取液提取后过滤,滤液经黄曲霉毒素免疫亲和柱净化,洗脱液经N2吹干后,加三氟乙酸和正己烷衍生后,用带有荧光检测器的超高效液相色谱仪测定。结果:四种黄曲霉毒素G2、G1、B2、B1得到很好的分离。检测限分别为:0.2μg/kg、0.4μg/kg、0.2μg/kg、0.4μg/kg。平均回收率在:81.5%~106.2%,相对标准偏差在1.1%~3.5%。结论:该法能使花生中黄曲霉毒素G2、G1、B2、B1得到很好的分离,结果准确。
OBJECTIVE: To determine aflatoxins in peanuts by using immunoaffinity column-high performance liquid chromatography. Methods: The sample was extracted with methanol + water (70:30), then filtrated. The filtrate was purified by aflatoxin immunoaffinity column. The eluate was air-dried with N2 and derivatized with trifluoroacetic acid and n-hexane. Fluorescence detector with ultra performance liquid chromatography. Results: Four aflatoxins G2, G1, B2, B1 were well separated. The detection limits were 0.2μg / kg, 0.4μg / kg, 0.2μg / kg and 0.4μg / kg, respectively. The average recoveries ranged from 81.5% to 106.2% with relative standard deviations between 1.1% and 3.5%. Conclusion: This method can make aflatoxin G2, G1, B2, B1 in peanut be well separated with accurate results.
