目的评估螺杆状幽门螺杆菌(Hp)及其球形体感染胃黏膜上皮细胞对CD44v6表达的影响。方法幽门螺杆菌及其球形体与胃黏膜上皮细胞共生培养,免疫组化、RT-PCR检测CD44v6蛋白表达。结果免疫组化及RT-PCR检测,螺杆状Hp及其球形体感染人胃黏膜上皮细胞GES1,诱导CD44v6的表达与上调,且螺杆状Hp与球形体诱导CD44v6存在差异有统计学意义(P<0.05)。结论螺杆状Hp及其球形体感染导致胃黏膜CD44v6表达增加,提示CD44v6在Hp相关性胃炎、胃癌的发生过程中起作用,Hp球形体在Hp治疗复发的流行病学具有重要的意义。
Objective To evaluate the effect of Helicobacter pylori (Helicobacter pylori) and its spheroids infected gastric epithelial cells on the expression of CD44v6. Methods Helicobacter pylori and its spheroids were co-cultured with gastric mucosal epithelial cells. The expression of CD44v6 protein was detected by immunohistochemistry and RT-PCR. Results Immunohistochemical and RT-PCR results showed that the infection of Helicobacter pylori (Hp) and its spheroid (GFP) induced the expression and up-regulation of CD44v6 in gastric mucosal epithelial cells, and there was a significant difference in the expression of CD44v6 between helical Hp and spheroids (P < 0.05). Conclusion The infection of Helicobacter pylori and its spheroid leads to the increase of CD44v6 expression in gastric mucosa, suggesting that CD44v6 plays a role in the pathogenesis of Hp-related gastritis and gastric cancer. The epidemiology of Hp spheroid recurrence in Hp therapy is of great significance.
