以葡萄皮脱分化组织为最初材料 ,用根癌农杆菌 70 2菌株感染 ,通过纸电泳证明胭脂碱合成醇基因的存在 ,并在无激素培养基上培养 5~ 6代以上 ,将此组织 (MS)作为本次实验的研究对象 .用 He- Ne激光辐照 ,对 MS进行诱变处理 ,通过继代选育 ,得到优良细胞系 T12 ,其特点是 :生长迅速 ,生长周期比对照组 (CK)缩短 1 0~ 1 5天 ;生长曲线的稳定期明显延长 ,白藜芦醇平均产量比 CK组提高 40 %.通过稳定性实验证明 T12 组遗传性状是可靠的 .
The grape skins dedifferentiated tissue as the initial material, with Agrobacterium tumefaciens 70 2 strains infected by paper electrophoresis to prove the presence of nopaline alkaloid gene and cultured in hormone-free medium for more than 5 to 6 generations, this tissue ( MS) as the object of this study.Using He-Ne laser irradiation, MS mutagenesis treatment, through the succession breeding, to obtain a good cell line T12, which is characterized by: rapid growth, growth cycle than the control group CK) was shortened from 10 days to 15 days, the stability of the growth curve was prolonged, and the average yield of resveratrol was increased by 40% than that of CK group. The stability of T12 group was proved by the stability test.
