采用水煎和醇提2种工艺,通过大鼠肠外翻模型评价黄连-大黄配伍药对中生物碱的体外肠吸收特征。以小檗碱、巴马汀、黄连碱、表小檗碱4种主要生物碱成分为代表,采用一测多评法对其进行定量分析,计算吸收速率常数,进而评价2种工艺中,4种生物碱成分在大鼠不同肠段的体外吸收特征。结果显示:2种不同工艺提取物中上述4种成分的高、中、低剂量在小肠段均为线性吸收,R~2大于0.99,符合零级吸收速率,小肠部位的吸收速率常数(K_a)表现为水煎组>醇提组。
