目的建立高效液相色谱-二极管阵列法同时测定发酵面制品中的柠檬黄、胭脂红、日落黄、诱惑红和亮蓝5种合成色素的方法。方法采用Kromasil C18色谱柱(250 mm×4.6 mm,10μm)。以甲醇+乙腈(1+1)和乙酸铵溶液(0.01 mol/L,pH=4.0)为流动相,梯度洗脱,流速1.0 ml/min,用二极管阵列检测器记录各个色谱峰的紫外吸收光谱,色谱检测波长254 nm。结果各色素在1.56~50.0μg/ml质量浓度范围内呈良好的线性关系,相关系数均为1.000 0。将该方法用于实际样品的检测,5种人工合成色素的加标回收率在98.9%~101.6%之间,相对标准偏差为0.11%~0.41%。结论该方法操作简便、灵敏、结果可靠。
OBJECTIVE To establish a method for the simultaneous determination of five synthetic pigments, lemon yellow, carmine, sunset yellow, seductive red and brilliant blue in fermented noodle products by high performance liquid chromatography-diode array. Methods Kromasil C18 column (250 mm × 4.6 mm, 10 μm) was used. The mobile phase consisted of methanol + acetonitrile (1 + 1) and ammonium acetate solution (0.01 mol / L, pH = 4.0) with gradient elution at a flow rate of 1.0 ml / min. The UV spectra of each chromatographic peak were recorded with a diode array detector , Detection wavelength 254 nm. Results The results showed that there was a good linear relationship between each pigment in the concentration range of 1.56 ~ 50.0μg / ml, the correlation coefficient was 1.000 0. The method was applied to the detection of real samples. The spiked recoveries of the five synthetic pigments ranged from 98.9% to 101.6% with relative standard deviations of 0.11% -0.41%. Conclusion The method is simple, sensitive and reliable.
