目的探讨糖尿病大鼠大血管组织蛋白酶S(Cat S)和核转录因子κB(NF-κB)的变化。方法25只雄性SD大鼠被随机分为对照组、模型组(1周,4周,8周,12周)。采用腹腔注射链脲霉素(STZ)方法复制大鼠糖尿病模型,HE染色观察主动脉管壁形态学变化,分别采用Western-blotting和RT-PCR方法检测大血管Cat S、NF-κB蛋白和mRNA的变化。结果造模后8周可见管壁结构的变化。造模后4周,Cat S蛋白和mRNA开始增加,8周达到高峰。NF-κB蛋白和mRNA在造模后1周开始增加,4周后达到表达高峰。结论Cat S和NF-κB在糖尿病大血管病变过程中起重要作用。
Objective To investigate the changes of cathepsin S and NF-κB in diabetic rats. Methods Twenty-five male SD rats were randomly divided into control group and model group (1 week, 4 weeks, 8 weeks and 12 weeks). The model of diabetic rats was established by intraperitoneal injection of streptozotocin (STZ). The morphological changes of the aortic wall were observed by HE staining. The expressions of Cat S, NF-κB and mRNA were detected by Western-blotting and RT-PCR respectively The change. Results 8 weeks after the model tube wall structure changes. Four weeks after model establishment, Cat S protein and mRNA began to increase and peaked at 8 weeks. NF-κB protein and mRNA began to increase 1 week after modeling, reached the peak expression after 4 weeks. Conclusion Cat S and NF-κB play an important role in the pathogenesis of diabetic macroangiopathy.
