东乡野生稻是迄今为止世界上耐冷性最强的稻种资源之一,研究耐冷性遗传特征,挖掘耐冷基因,对把耐冷优良特性应用到水稻育种中具有重要意义。本研究结合前人对东乡野生稻耐冷性定位结果,以东乡野生稻为供体亲本,协青早B(XQZB)为受体亲本,结合分子标记,回交构建近等基因系,再用耐冷近等基因系与协青早B杂交加自交,得到F2群体作为试验材料,对耐冷主效QTL-qCT10进行分离鉴定。最后把qCT10界定在第10染色体分子标记In Del10-9与InDel10-22间224.5 kb范围内,是一个新主效耐冷QTL。研究结果有助于进一步探明东乡野生稻强耐冷性遗传机制,为培育强耐冷水稻品种打下基础。
Dongxiang Wild Rice (Oryza sativa L.) is one of the most cold tolerant rice resources in the world so far. Studying the genetic characters of cold tolerance and digging cold tolerance genes are of great significance for applying excellent cold tolerance characteristics to rice breeding. In this study, based on the results of previous studies on the cold tolerance of Dongxiang wild rice, we used the Dongxiang wild rice as the donor parent and XQZB as the recipient parent to construct a near-isogenic line combining with molecular markers and backcrossing, Near-isogenic lines were crossed with Xieqingzao B to obtain F2 populations as test materials, and the QTLs for cold-tolerance QTLs-qCT10 were isolated and identified. Finally, qCT10 was defined as a new QTL for major cold-tolerance in the range of 224.5 kb between In Del 10-9 and InDel 10-22. The results will help further explore the genetic mechanism of strong cold tolerance in Dongxiang wild rice and lay a foundation for cultivating strong cold-tolerant rice varieties.
