目的研究蜥蜴胃康方及其拆方含药SD大鼠血清对人胃癌细胞BCG-823增殖凋亡,以及细胞E-钙黏素(Ecad)、整合素(Integrin)、基质金属蛋白酶(MMP-9)蛋白表达水平的影响,阐明蜥蜴胃康方干预胃癌细胞的作用及其机制。方法 70只SPF级雄性SD大鼠,随机分为7组:生理盐水对照组(A),蜥蜴胃康方组(B)、拆方补气组(C)、养阴组(D)、解毒通络组(E)、华蟾素片对照组(F)、5-FU对照组(G),每组10只。各组分别用相应浓度药物灌胃1周后处死,于腹主动脉处取血、离心,制备含药血清,加于人胃癌细胞BCG-823培养48h,MTT法检测含药血清对胃癌细胞增殖凋亡的影响;Western-Blot检测胃癌细胞Integrin、E-cad、MMP-9蛋白表达的变化。结果各含药大鼠血清干预组OD均值小于生理盐水对照组;蜥蜴胃康方组(B)及其拆方C、E组与对照组F、G比较,P<0.05;蜥蜴胃康方组B与拆方组C、D、E比较,P<0.05;拆方组C、D、E组之间比较,P<0.05,以补气组C作用较佳,其次为解毒通络组;Integrin、E-cad蛋白表达与生理盐水组比较,呈显著升高趋势;MMP-9蛋白表达与生理盐水组比较则显著降低,均以蜥蜴胃康方组为著。结论蜥蜴胃康方可通过影响Integrin、E-cad及MMP-9蛋白表达,抑制细胞增殖,促进细胞凋亡,具有较好的抗癌作用。
Objective To study the proliferation and apoptosis of human gastric cancer cell line BCG-823 induced by Lizhenweikang decoction and its subgroups of drug-containing SD rat serum, 9) protein expression level, to clarify the lizard stomach Kang intervention of gastric cancer cells and its mechanism. The rats in each group were sacrificed one week after gavage with the corresponding drug, blood was collected from the abdominal aorta and centrifuged to prepare medicated serum. The serum was added to human gastric cancer cell line BCG-823 for 48 hours. MTT assay was used to detect the proliferation of gastric cancer cells Apoptosis; Western-Blot detection of gastric cancer cells Integrin, E-cad, MMP-9 protein expression changes. Results The OD mean value of the serum-containing intervention group was lower than that of the saline control group (P <0.05); lizard Weikangfang group (B) and its disassembled group C and E were compared with those of the control group B and C, D, E, P <0.05; C, D, E groups were compared, P <0.05, with qi group C is better, followed by Jiedu Tongluo group; Integrin , E-cad protein expression was significantly increased compared with the saline group; MMP-9 protein expression was significantly reduced compared with the saline group, both lizards Weikangfang group. Conclusion Lizards Weikang formula can inhibit the expression of Integrin, E-cad and MMP-9, inhibit cell proliferation, promote apoptosis, and has good anticancer effect.
