目的探讨星形细胞上调基因-1(astrocyte elevated gene-1,AEG-1)在原发性肝细胞癌组织中的表达及其意义。方法 60例肝细胞癌患者,取其肝癌组织60份为肝癌组,癌旁肝组织54份为对照组,采用荧光定量PCR法检测2组AEG-1mRNA表达情况,采用Western blot法检测AEG-1蛋白表达情况,采用免疫组织化学法检测AEG-1阳性表达率,分析AEG-1蛋白表达与病理指标间的关系。结果肝癌组AEG-1mRNA和蛋白相对表达量(4.71±2.87、1.27±0.62)及AEG-1阳性表达率(53.3%)均明显高于对照组(1.43±0.49、0.69±0.25、29.6%)(P<0.05);肝癌组织AEG-1阳性表达水平在肿瘤转移及病理分级上差异无统计学意义(P>0.05)。结论 AEG-1在原发性肝细胞癌组织中特异性表达增高,可能是诊断原发性肝细胞癌的生物学标记物。
Objective To investigate the expression of astrocyte elevated gene-1 (AEG-1) in primary hepatocellular carcinoma (HCC) and its significance. Methods 60 patients with hepatocellular carcinoma (HCC) were divided into two groups: liver cancer group (60) and liver cancer group (54) as the control group. The expression of AEG-1 mRNA in the two groups was detected by fluorescence quantitative PCR. Western blot was used to detect AEG-1 The protein expression of AEG-1 was detected by immunohistochemical method. The relationship between the expression of AEG-1 and pathological parameters was analyzed. Results The relative expression of AEG-1mRNA and protein (4.71 ± 2.87,1.27 ± 0.62) and AEG-1 positive rate (53.3%) were significantly higher in hepatocellular carcinoma group than in control group (1.43 ± 0.49,0.69 ± 0.25,29.6%) P <0.05). The positive expression level of AEG-1 in HCC tissues showed no significant difference between tumor metastasis and pathological grade (P> 0.05). Conclusion AEG-1 is highly expressed in primary hepatocellular carcinoma and may be a biomarker for the diagnosis of primary hepatocellular carcinoma.
