目的:测定我国恶性疟原虫环子孢子蛋白部分基因序列,了解我国虫株与其它虫株CS蛋白基因序列的差异。方法:采用PCR方法特异性扩增CS蛋白基因片段,并将该基因片段克隆于M13噬菌体,取3个阳性克隆制备M13-CSP单链DNA,用双脱氧链末端终止法测定该基因片段核苷酸序列。应用PCGENE软件对六个虫株CS蛋白基因序列进行了一系列比较分析。结果:我国恶性疟原虫云南株(PFD-3/YN)与T4、Welcome、NF54、3D7及7G8等虫株CS蛋白基因序列有不同程度的差异,其中一个有意义核苷酸突变发生在P.fTh/Tc抗原表位区。结论:云南株与其它虫株CS蛋白基因存在差异。
Objective: To determine the gene sequence of circumsporozoite protein of Plasmodium falciparum in our country, and to understand the difference of the sequence of CS protein gene in China and other insect strains. Methods: The CS gene fragment was amplified by PCR and cloned into M13 phage. Three positive clones were used to prepare M13-CSP single-stranded DNA. The nucleotide sequence of the fragment was determined by dideoxy chain termination Acid sequence. A series of comparative analyzes of the CS protein gene sequences of six insect strains were carried out using PCGENE software. Results: The sequence of CS protein of P. falciparum Yunnan strain (PFD-3 / YN) was different from that of T4, Welcome, NF54, 3D7 and 7G8 strains. One of the nucleotide mutations in P. falciparum occurred in P. fTh / Tc epitope region. Conclusion: There are differences in CS protein gene between Yunnan and other strains.
