目的探讨去坐骨神经方法建立幼鼠废用性骨质疏松动物模型,为研究生长期骨骼发生的废用性骨质疏松提供理论借鉴。方法20只4周龄SD雄性幼年大白鼠,随机分为2组行手术,8周后,将模型组(SN)及假手术组(SHAN组)比较,采用骨矿含量、骨密度测量、骨组织病理学及骨形态计量学指标、骨生物力学测量对幼鼠废用性骨质疏松动物模型进行综合评价。结果假手术组大白鼠左侧胫骨骨矿含量、骨密度、骨小梁体积密度、股骨几何参数以及结构力学性能皆不同程度高于模型组。结论采用去坐骨神经的方法可成功建立幼鼠废用性骨质疏松动物模型。
Objective To explore the method of sciatic nerve to establish an animal model of disuse osteoporosis in young rats and to provide a theoretical reference for the study of disuse osteoporosis in growing skeletal bone. Methods Twenty male Sprague-Dawley (SD) rats of 4 weeks old were randomly divided into 2 groups. After 8 weeks, the model group (SN) and sham operation group (SHAN group) were compared. Bone mineral density, Histopathology and bone morphometry, bone biomechanical measurements of animal models of obesity in young rats a comprehensive evaluation. Results The left tibia bone mineral content, bone mineral density, trabecular bone volume density, femoral geometric parameters and mechanical properties of the sham group were all higher than those in the model group. Conclusion The sciatic nerve can be successfully used to establish an animal model of disuse osteoporosis in young rats.
