目的比较血浆循环DNA(circulating DNA,cDNA)在原发性干燥综合征(primary Sjgren’s syndrome,pSS)和淋巴瘤(Lymphomas)中的差异。方法选用人类基因组管家基因GAPDH设计引物,测定pSS、淋巴瘤及正常对照组中血浆cDNA的值,同时测定血沉(ESR)、C-反应蛋白(CRP)、免疫球蛋白(IgA、IgG、IgM),并评定pSS的活动指数(Sjgren’s syndrome disease activity index,SSDAI)和损害指数(Sjgren’s syndrome disease damage index,SSDDI)、淋巴瘤国际预后指标(Lymphoma international prognostic index,IPI),分析cDNA与相关参数之间的关系。结果 3组间血浆cDNA两两比较:pSS组与淋巴瘤组、正常对照组比较差异均有统计学意义(P<0.05);pSS组中:cDNA浓度值与IgG呈正相关(r=0.333,P<0.05),cDNA浓度值与SSDAI呈正相关(r=0.551,P<0.01),cDNA浓度值与SSDDI无相关性(r=0.198,P>0.05),淋巴瘤组cDNA浓度值与IPI正相关(r=0.596,P<0.01);pSS组IgA、IgG均明显高于淋巴瘤组(P<0.05)。结论血浆cDNA在pSS及淋巴瘤中均升高,且两者之间有差异,作为pSS与淋巴瘤疾病鉴别的一种新的生物学标记,也可作为淋巴瘤预后及pSS疾病活动的观察指标,或许还可作为pSS向淋巴瘤发展的监测指标之一。
Objective To compare the differences of circulating DNA (cDNA) in primary Sjögren’s syndrome (pSS) and lymphoma (Lymphomas). Methods The human genomic housekeeping gene GAPDH was used to design the primers for the determination of plasma cDNA in pSS, lymphoma and normal control group. ESR, C-reactive protein (CRP), immunoglobulin (IgA, IgG, IgM) (SSDII) and Sjögren’s syndrome disease damage index (SSDDI), Lymphoma international prognostic index (IPI) were analyzed. The results of cDNA and The relationship between the relevant parameters. Results There was a significant difference in plasma cDNA between the three groups (p <0.05). There was a significant difference between the pSS group and the lymphoma group (P <0.05) (R = 0.198, P> 0.05). There was a positive correlation between the concentration of cDNA and SSDAI (r = 0.551, P <0.01) r = 0.596, P <0.01). IgA and IgG in pSS group were significantly higher than those in lymphoma group (P <0.05). Conclusions Plasma cDNA is elevated in both pSS and lymphoma, and there is a difference between the two. As a new biomarker for the identification of pSS and lymphoma, plasma cDNA can also be used as an indicator of lymphoma prognosis and pSS disease activity , May also serve as one of the indicators of the development of pSS to lymphoma.
