本研究用组织化学法、扫描和透射电镜观察了人胎儿胸腺哺育细胞(thymic nurse cell,TNC)。结果表明,TNC 的 ANAE、AcP、AIP、ATPase、5′-Nase 和 PAS 均呈阴性反应,TNC胞质内含有数量不等的淋巴细胞,其中少数淋巴细胞的 ANAE、AcP、AIP 和 ATPase 呈阳性反应。TNC 内的淋巴细胞 ANAE 阳性率为26.8%。TNC 外的淋巴细胞 ANAE 阳性率为51.3%,两者的结果比较,有显著性差异(P<0.01,u=11.23)。扫描电镜观察,TNC 为球形,细胞表面较光滑,有少量短微绒毛。1μm 半薄切片显示,胸腺皮质内可见 TNC 样细胞,细胞界限清楚,核圆或椭圆形,染色淡。胞质中可见数量不等的淋巴细胞。透射电镜观察,TNC 胞质内可见张力丝,表明这些细胞具有上皮细胞特征。TNC 胞质内的淋巴细胞可见正常的线粒体、游离核糖体等细胞器。本文对上述结果与胸腺巨噬细胞作了比较和讨论。
In this study, human fetal thymic nurse cells (TNC) were observed by histochemistry, scanning and transmission electron microscopy. The results showed that ANAE, AcP, AIP, ATPase, 5’-Nase and PAS in TNC all showed negative reaction. TNC cytoplasm contained a varying number of lymphocytes. ANAE, AcP, AIP and ATPase of a few lymphocytes were positive reaction. The positive rate of lymphocytic ANAE in TNC was 26.8%. The positive rate of ANAE of lymphocytes outside TNC was 51.3%, there was a significant difference between the two (P <0.01, u = 11.23). Scanning electron microscopy, TNC spherical, cell surface is smooth, a small amount of short microvilli. Semi-thin sections of 1μm showed that TNC-like cells were seen in the thymic cortex, with clear cell boundaries, nucleus or oval, and light staining. Cytoplasm can be seen in varying numbers of lymphocytes. Transmission electron microscopy showed that there were tension filaments in the cytoplasm of TNC, indicating that these cells have epithelial cell features. TNC cytoplasmic lymphocytes can be seen normal mitochondria, free ribosomes and other organelles. This article compares and discusses the above results with thymus macrophages.
