目的:建立同时测定牛黄清感胶囊中绿原酸、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、连翘苷6个成分的方法。方法:采用Platisil ODS色谱柱(250 mm×4.6 mm,5μm),柱温30℃,以乙腈(A)-0.1%磷酸水溶液(B)为流动相,梯度洗脱(0~20 min,10%A→16%A;20~40 min,16%A→30%A;40~60 min,30%A→40%A;60~80 min,40%A),流速1mL·min~(-1),变换波长检测(278 nm处检测绿原酸、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素;210 nm处检测连翘苷)。结果:绿原酸、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、连翘苷的线性范围分别为0.016 4~0.328μg(r=0.999 6)、0.106~2.12μg(r=0.999 6)、0.042~0.84μg(r=0.999 7)、0.026 3~0.526μg(r=0.999 8)、0.015~0.3μg(r=0.999 4)、0.033 3~0.666μg(r=0.999 4),平均回收率(n=9)分别为99.6%(RSD=1.2%)、99.3%(RSD=1.1%)、100.7%(RSD=1.4%)、99.6%(RSD=1.2%)、100.2%(RSD=1.5%)、99.6%(RSD=1.8%)。3批样品中绿原酸、黄芩苷、汉黄芩苷、黄芩素、汉黄芩素、连翘苷的含量分别在0.755~0.762、34.218~34.230、1.401~1.404、0.774~0.788、0.511~0.522、0.732~0.740 mg·g~(-1)。结论:经方法学验证,本法可作为牛黄清感胶囊质量控制的一个有效的方法。
Objective: To establish a method for the simultaneous determination of 6 components of chlorogenic acid, baicalin, baicalin, baicalein, wogonin and forsythin in Niuhuang Qingyan Capsule. Methods: Platisil ODS column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile (A) -0.1% phosphoric acid solution (B) A → 16% A; 20-40 min, 16% A → 30% A; 40-60 min, 30% A → 40% A; 60-80 min, 40% ), Wavelength detection (detection of chlorogenic acid, baicalin, baicalin, baicalein and wogonin at 278 nm; detection of forsythin at 210 nm). Results: The linear range of chlorogenic acid, baicalin, baicalin, baicalein, wogonin and forsythin were 0.016 4 ~ 0.328μg (r = 0.999 6) and 0.106 ~ 2.12μg (r = 0.999 6) , 0.042-0.84μg (r = 0.999 7), 0.026-3-0.526μg (r = 0.999 8), 0.015-0.3μg (r = 0.999 4), 0.033 3- 0.666μg (r = 0.999 4) .The average recoveries 99.7% (RSD = 1.4%), 99.6% (RSD = 1.2%), 100.2% (RSD = 1.5% ), 99.6% (RSD = 1.8%). The content of chlorogenic acid, baicalin, baicalin, baicalein, wogonin and forsythin in three batches of samples were respectively in the range of 0.755-0.762, 34.218-34.230, 1.401-1.404, 0.774-0.788, 0.511-0.522, 0.732 ~ 0.740 mg · g -1. Conclusion: This method can be used as an effective method for the quality control of Niuhuang Qinggan Capsule by methodological verification.
