目的探讨梯度离心法对不同质量精液的影响,分析其最佳离心力与离心时间。方法收集不同质量精液分组处理,在不同的离心力和离心时间作用下对各组标本进行梯度离心处理。离心处理后分析各组在不同离心力和离心时间的作用下收集到的精子质量,比较各组的精子密度与活力。结果对于高质量的精液标本最佳离心力与离心时间是200g离心10~15min;普通质量的精液是300g下离心15~20min;对于低质量精液即使加大离心力,增加离心时间也无法达到预期的离心效果。结论在人类辅助生殖技术(ART)中,相同离心力与离心时间对不同质量的精液处理效果不同,不同离心力离心时间下对相同质量精液的处理效果也有差别。
Objective To investigate the effect of gradient centrifugation on different quality semen and analyze its optimal centrifugal force and centrifugation time. Methods Different concentrations of seminal fluid were collected and treated by gradient centrifugation under different centrifugal force and centrifugation time. After centrifugation, the quality of sperm collected under different centrifugal force and centrifugation time was analyzed. The sperm density and vitality of each group were compared. Results The optimal centrifugal force and centrifugation time for high-quality semen samples were 200g centrifuged for 10 ~ 15min; normal quality semen was centrifuged at 300g for 15 ~ 20min; for low quality semen, even if the centrifugal force was increased, the centrifugation time could not reach the expected centrifugation effect. Conclusions In human assisted reproductive technology (ART), the same centrifugal force and centrifugation time have different effects on the quality of semen, and the treatment effect of the same quality semen is different under different centrifugal force centrifugation time.
