TILLING技术是一种高效检测SNP的反向遗传学方法。根据已克隆的水稻耐盐主效基因SKC1设计特异引物,以化学诱变剂EMS(甲基磺酸乙酯)处理获得的1 310份M2代水稻为材料,利用TILLING技术检测突变位点。经过2011~2012年2年M2、M3代的连续检测及测序分析,获得SKC1基因纯合突变体5份,杂合突变体14份,研究结果表明利用TILLING技术进行水稻耐盐突变体筛选是非常有效的。
TILLING technology is a reverse genetics approach that efficiently detects SNPs. Based on the cloned rice SKC1 salt-tolerant major gene design specific primers, 1 310 M2 generation rice obtained by chemical mutagenesis EMS (methyl methanesulfonate) were used as materials to detect the mutation sites by TILLING technique.
